Biological Background: LATS2 Function and Localization
- LATS2 (serine/threonine-protein kinase LATS2) is also known as kinase phosphorylated during mitosis protein (KPM), large tumor suppressor homolog 2, and warts-like kinase. It is a core component of the Hippo signaling pathway.
- LATS2 negatively regulates YAP1 by phosphorylating it and inhibiting its nuclear translocation. This action restricts proliferation and promotes apoptosis, acting as a key tumor suppressor. Related references: PMID:18158288 PMID:26437443 PMID:26598551 PMID:34404733
- Within the Hippo kinase cascade, MST1/2 (STK4/3) in complex with SAV1 phosphorylates and activates LATS2 in complex with MOB1. Activated LATS2 then phosphorylates YAP1 and WWTR1/TAZ. Related references: PMID:26437443 PMID:26598551 PMID:34404733

- Subcellularly, LATS2 localizes to the cytoplasm, centrosome, spindle poles, and nucleus.
- Tissue specificity: LATS2 is expressed at high levels in heart and skeletal muscle, with lower expression in other tissues.
