Biological Background: LATS1 Function and Localization
- LATS1 (Large tumor suppressor homolog 1), also known as WARTS, is a serine/threonine-protein kinase encoded by the LATS1 gene and belongs to the Hippo signaling pathway.
- Phosphorylation of YAP1 by LATS1 inhibits its nuclear translocation, suppressing cell proliferation and promoting apoptosis, thereby controlling organ size and tumor suppression. Related references: PMID:10518011, PMID:10831611, PMID:18158288
- The core kinase cascade involves STK3/MST2 and STK4/MST1 in complex with SAV1, which phosphorylate LATS1/2 in complex with MOB1, leading to phosphorylation and inactivation of YAP1 and WWTR1/TAZ. Related references: PMID:18158288, PMID:26437443, PMID:28068668
- Acts as a tumor suppressor maintaining ploidy through regulation of mitotic progression and the G1 tetraploidy checkpoint. Related references: PMID:15122335, PMID:19927127
- Negatively regulates G2/M transition by down-regulating CDK1 kinase activity. Related references: PMID:9988268
- Affects cytokinesis by regulating actin polymerization via negative modulation of LIMK1. Related references: PMID:15220930
- Plays a role in mammary gland epithelial cell differentiation through the Hippo pathway and by promoting degradation of ESR1. Related references: PMID:28068668
- Activates the NLRP3 inflammasome by mediating phosphorylation of NLRP3 at Ser-265, enabling NLRP3 activation by NEK7. Related references: PMID:39173637
- Subcellular localization includes centrosome, spindle, midbody, and spindle pole body, consistent with its roles in mitosis and cytokinesis.
- Expressed ubiquitously in adult tissues except lung and kidney.
Experimental Guidance and Technical Tips
- For Western blot (WB), note the predicted molecular weight of LATS1 is ~127 kDa. Consider using a low percent SDS-PAGE gel (e.g., 6–8%) and appropriate transfer conditions for high-molecular-weight proteins.
- In immunohistochemistry (IHC-P), antigen retrieval may be required; validate the optimal method in your tissue samples. Positive control tissues should be selected based on known expression (e.g., tissues other than lung or kidney).
- For ELISA, the immunogen (synthetic peptide corresponding to amino acids 100–200) may serve as a positive control. Include appropriate negative controls to assess specificity.
- As a polyclonal antibody, batch-to-batch consistency should be confirmed for critical assays. Consider validating reactivity in the relevant species (human, mouse, rat) and sample types.
