Biological Background: LATS1 Function and Localization

• LATS1 (Large tumor suppressor homolog 1), also known as WARTS protein kinase (h-warts), is a serine/threonine-protein kinase encoded by the LATS1 gene on chromosome 6. It is a key tumor suppressor and component of the Hippo signaling pathway.
• As a core Hippo pathway component, LATS1 restricts cell proliferation and promotes apoptosis. It forms a kinase cascade with STK3/MST2 and STK4/MST1, phosphorylates and inactivates YAP1 and TAZ, preventing their nuclear translocation and oncogenic transcription.
