Biological Background: LAT Function and Localization

  • LAT (Linker for activation of T-cells family member 1), also known as pp36 or p36-38, is a 36 kDa transmembrane adapter protein essential for T-cell receptor (TCR) and pre-TCR signaling in mature T-cells and during T-cell development. Related references: PMID:23514740, PMID:25907557
  • It couples receptor activation to downstream signaling events, including calcium mobilization, PKC activation, MAPK signaling, and cytoskeletal reorganization, by recruiting adaptor proteins such as GRB2, GRAP2, and PLCG1.
  • LAT is involved in FCGR3-mediated signaling in natural killer (NK) cells and FCER1-mediated signaling in mast cells, highlighting its role in innate immune responses.
  • The protein is localized to the cell membrane via a signal-anchor transmembrane helix and undergoes post-translational modifications including palmitoylation, phosphorylation, and ubiquitination.
  • Tissue expression is predominantly in thymus, T-cells, NK cells, and mast cells, with lower levels in spleen; it is present in T-cells but not B-cells at the protein level.

  • The immunogen (amino acids 95-262) covers the cytoplasmic domain of LAT, including critical tyrosine phosphorylation sites; the antibody may detect both phosphorylated and non-phosphorylated forms.
  • For Western blot, expect a band around 28-36 kDa (supplier reports 28 kDa; UniProt theoretical weight 27.9 kDa; phospho-p36 forms may run higher). Validate in a relevant cell line such as Jurkat (T-cell) with appropriate controls.
  • ELISA usage should be optimized with recombinant or native LAT; consider coating with the immunogen region for capture assays.
  • As a rabbit polyclonal antibody, lot-to-lot consistency may vary; we recommend titrating and validating each new lot in your specific assay conditions.
  • Cross-reactivity has been demonstrated for human LAT; for other species, perform sequence alignment and experimental validation.
