Biological Background: LARP1 Function and Localization
- LARP1 (La-related protein 1) is an RNA-binding protein that regulates the translation of specific target mRNAs downstream of the mTORC1 complex in response to growth signals and nutrient availability. Related references: PMID:20430826, PMID:23711370, PMID:24532714
- It interacts with both the 3' poly-A tails and the 5' terminal oligopyrimidine (5'TOP) motifs of mRNAs, repressing translation of 5'TOP-containing transcripts by preventing EIF4G1 binding. Related references: PMID:23711370, PMID:25940091, PMID:26206669, PMID:28379136, PMID:28650797
- Upon mTORC1 activation, LARP1 is phosphorylated and dissociates from the 5' untranslated region (UTR), relieving translational repression. Related references: PMID:25940091, PMID:28650797
- Under normal nutrient availability, LARP1 primarily associates with the 3' UTR of ribosomal protein mRNAs and stimulates their translation, associating with actively translating ribosomes. Related references: PMID:23711370, PMID:28650797
- LARP1 stabilizes mRNAs containing a 5'TOP motif, a process required to prevent apoptosis. Related references: PMID:20430826, PMID:23711370, PMID:25940091
- It localizes predominantly to the cytoplasm and can be found in cytoplasmic granules.
- Positively regulates the replication of dengue virus (DENV) upon infection.
- Keywords: Acetylation, Alternative splicing, Direct protein sequencing, Isopeptide bond, Methylation, Phosphoprotein, Protein biosynthesis, Repressor, RNA-binding, Translation regulation, Ubl conjugation.
Experimental Guidance and Technical Tips
- For Western blotting, given the large predicted molecular weight (124 kDa), consider using a low-percentage SDS-PAGE gel (e.g., 6–8%) and extended transfer times to ensure efficient detection.
- In ELISA applications, the recombinant immunogen (amino acids 106–205) may serve as a useful positive control for antigen-coating optimization.
- As the antibody is raised against a human-specific immunogen, validate reactivity in the relevant human cell lines or tissue lysates and consider testing cross-reactivity with other species if required.
- Because LARP1 is a phosphoprotein regulated by mTORC1 signaling, ensure that cell culture conditions (serum levels, nutrient status) are controlled to interpret expression and modification patterns accurately.
