Biological Background: LAPTM4B Function and Localization

  • LAPTM4B, also known as lysosomal-associated transmembrane protein 4-beta (LAPTM4β) and LC27, is encoded by the LAPTM4B gene in humans.
  • Required for optimal lysosomal function. Related references: PMID:21224396
  • Blocks EGF-stimulated EGFR intraluminal sorting and degradation; regulates PIP5K1C interaction and inhibits HGS ubiquitination. Related references: PMID:25588945
  • Recruits the leucine transporter complex SLC3A2-SLC7A5 to lysosomes, promoting essential amino acid entry and activating V-ATPase and mTORC1 signaling. Related references: PMID:25998567
  • Functions as a negative regulator of TGFB1 production in regulatory T cells. Related references: PMID:26126825
  • Binds ceramide and facilitates its exit from late endosomes, thereby controlling cell death pathways. Related references: PMID:26280656
  • Localizes to the endomembrane system, including late endosome, lysosome, multivesicular body, and cell membrane, and is found in cell projections.
  • Annotated with keywords including alternative splicing, transmembrane helix, transport, and ubiquitin conjugation.

Experimental Guidance and Technical Tips

  • The immunogen was designed using a recombinant fusion protein corresponding to the cytosolic region of human LAPTM4B (amino acids 218-317), which may provide specificity for detecting the full-length protein.
  • For Western blot (WB), consider using a dilution range of 1:500–1:2000 and validate in lysates that endogenously express LAPTM4B, such as lysosome-enriched fractions or cancer cell lines known to upregulate LAPTM4B.
  • For ELISA, titration is recommended to determine the optimal coating concentration and detection range; include proper controls for nonspecific binding.
  • As the antibody cross-reacts with mouse and rat LAPTM4B, it may be suitable for interspecies studies; however, verify target sequence homology in the immunogen region.
