Biological Background: LAMP2 Function and Localization
- LAMP2 (Lysosome-associated membrane glycoprotein 2), also known as CD107b, LAMPB, or LGP-96, is encoded by the LAMP2 gene and exists as three alternatively spliced isoforms: LAMP-2A, LAMP-2B, and LAMP-2C.
- Essential for lysosome biogenesis and maintenance of lysosomal pH by directly inhibiting the proton channel TMEM175, thereby facilitating lysosomal acidification for optimal hydrolase activity (PubMed:37390818).
Related references: PMID:37390818 - Plays a critical role in chaperone-mediated autophagy (CMA) by binding substrate proteins (e.g., GAPDH, GPX4, NLRP3, MLLT11) recognized by HSPA8/HSC70 and targeting them for lysosomal degradation, a process important for protein quality control and stress responses (PubMed:11082038, PubMed:18644871, PubMed:24880125, PubMed:36586411).
Related references: PMID:11082038 PMID:18644871 PMID:24880125 PMID:36586411 - Required for the fusion of autophagosomes with lysosomes during macroautophagy, as cells lacking LAMP2 fail to accumulate STX17 on autophagosomes, preventing fusion and degradation of autophagic contents (PubMed:27628032).
Related references: PMID:27628032 - Facilitates efficient MHC class II-mediated presentation of exogenous antigens by ensuring proper lysosomal degradation, although it is not required for endogenous antigen presentation (PubMed:15894275, PubMed:20518820).
Related references: PMID:15894275 PMID:20518820 - Localizes to the membranes of lysosomes, late endosomes, autophagosomes, and the cell surface, consistent with its trafficking through the endocytic and autophagic pathways.
- Isoforms show tissue-specific expression: LAMP-2A is high in placenta, lung, and liver; LAMP-2B is broadly expressed; LAMP-2C is detected in intestine, heart, brain, and skeletal muscle (PubMed:26856698, PubMed:7488019).
Related references: PMID:26856698 PMID:7488019 - Heavily glycosylated with disulfide bonds; classified as a glycoprotein and transmembrane protein. Associated with Danon disease (glycogen storage disease due to LAMP2 deficiency).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the luminal domain of human LAMP2 (aa 29-375), which is highly glycosylated. Consider this when interpreting molecular weight shifts in western blot; the observed band may appear larger than the predicted 45 kDa due to extensive glycosylation.
- For western blot (WB), validate the antibody in your specific sample matrix; use reducing conditions and load appropriate controls.
- For immunohistochemistry (IHC-P), antigen retrieval methods (e.g., heat-induced epitope retrieval) may be required to unmask LAMP2 epitopes, especially in formalin-fixed paraffin-embedded tissues.
- In immunofluorescence (IF/ICC), LAMP2 staining should show punctate lysosomal pattern; co-staining with lysosomal markers (e.g., LAMP1) is recommended for confirmation.
- This polyclonal antibody may exhibit batch-to-batch variation; include positive and negative controls in each experiment.
