Biological Background: LAMP1/CD107a Function and Localization
- LAMP1/CD107a is a heavily glycosylated lysosomal membrane glycoprotein with a predicted unglycosylated molecular weight of 43 kDa; the mature protein migrates at ~120 kDa (LGP-120).
- It is essential for lysosome biogenesis, lysosomal pH regulation, autophagy, and cholesterol homeostasis (PubMed:15121881). Related references: PMID:15121881
- In NK cells, LAMP1/CD107a regulates cytotoxicity by mediating granule trafficking and perforin delivery, and it protects NK cells from autolytic damage (PubMed:23847195). Related references: PMID:23847195
- The protein promotes tumor cell metastasis, possibly by interacting with selectins on the cell surface (PubMed:2676155). Related references: PMID:2676155
- LAMP1/CD107a localizes predominantly to the membranes of late endosomes and lysosomes, and trans-locates to the cell surface and cytolytic granule membranes during cellular activation.
- Structurally, it comprises a signal peptide, a large lumenal domain with multiple N-glycosylation sites, a single transmembrane helix, and a short cytoplasmic tail, consistent with its role as a stable membrane anchor.
Experimental Guidance and Technical Tips
- The antibody is raised against recombinant full-length or partial human/mouse CD107a, ensuring recognition of the native and denatured protein in both human and mouse samples.
- For Western blotting (WB), the reduced form of LAMP1/CD107a migrates at approximately 43 kDa; for the heavily glycosylated mature form, non-reducing conditions may reveal a 120 kDa band.
- IHC-P and IF/ICC detection of this lysosomal membrane protein may require permeabilization (e.g., 0.1% Triton X-100) and optimized fixation for best signal-to-noise ratio.
- ELISA performance can be validated by coating recombinant protein or native cell lysates and titrating the antibody under standard indirect ELISA conditions.
- As a KD-validated clone, it is suitable for quantitative analysis and may be combined with knockout controls to confirm specificity.
