Biological Background: LAMP1/CD107a Function and Localization

  • Lysosome-associated membrane glycoprotein 1 (LAMP1), also known as CD107a or LAMPA, is a heavily glycosylated type I transmembrane protein encoded by the LAMP1 gene. Alternative splicing generates multiple isoforms.
  • Functions in lysosome biogenesis, pH regulation, autophagy, and cholesterol homeostasis by inhibiting the TMEM175 proton channel, thus facilitating lysosomal acidification (PubMed:37390818). Related references: PMID:37390818
  • Key player in natural killer (NK) cell cytotoxicity: essential for cytotoxic granule movement and perforin trafficking to the lytic granule, and protects NK cells from self-inflicted damage (PubMed:23632890, PubMed:23847195, PubMed:2022921). Related references: PMID:23632890 PMID:23847195 PMID:2022921
  • Serves as a receptor for Lassa virus glycoprotein, promoting viral entry, and also supports FURIN-mediated cleavage of mumps virus fusion protein F (PubMed:24970085, PubMed:25972533, PubMed:27605678, PubMed:29295909). Related references: PMID:24970085 PMID:25972533 PMID:27605678 PMID:29295909
  • Presents carbohydrate ligands to selectins, mediating cell adhesion (PubMed:7685349). Related references: PMID:7685349
  • Predominantly localized to lysosomal and late endosomal membranes, but can also traffic to the cell surface and cytolytic granule membranes under certain conditions.
  • Contains a large lumenal domain with multiple N-linked glycosylation sites and disulfide bonds, a single-pass transmembrane helix, and a short cytoplasmic tail.

Experimental Guidance and Technical Tips

  • The immunogen used to generate this antibody corresponds to amino acids 29-382 of human LAMP1, encompassing the entire lumenal domain. Consider this when designing epitope-based assays.
  • For Western blot (WB), LAMP1 appears as a diffuse band due to heavy glycosylation, with a predicted molecular weight of ~45 kDa (unglycosylated precursor ~38 kDa). Optimal detection may require careful sample preparation and adequate blocking.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization steps may be necessary to access the lysosomal compartment.
  • For flow cytometry (FC), intracellular staining protocol is recommended to detect total LAMP1, while cell surface staining can visualize LAMP1 translocated to the plasma membrane upon activation (e.g., degranulation assay).
  • This antibody is suitable for ELISA; validate pairing with appropriate capture/detection reagents in the relevant sample matrix.
