Biological Background: LAMP1/CD107a Function and Localization
- LAMP1/CD107a (Lysosome-associated membrane glycoprotein 1) is a heavily glycosylated type I transmembrane protein and a major component of the lysosomal membrane, also known as LGP-120 or CD107a.
- Plays a critical role in lysosome biogenesis and pH regulation by acting as a direct inhibitor of the proton channel TMEM175, facilitating lysosomal acidification (By similarity).
- Essential for autophagy and cholesterol homeostasis; LAMP1 deficiency impairs autophagic flux and leads to cholesterol accumulation (PubMed:15121881). Related references: PMID:15121881
- Involved in natural killer (NK) cell cytotoxicity by promoting cytotoxic granule movement to the cell surface and perforin trafficking, while also protecting NK cells from self-damage during degranulation (PubMed:23847195). Related references: PMID:23847195
- Presents carbohydrate ligands to selectins and is implicated in tumor cell metastasis (PubMed:2676155). Related references: PMID:2676155
- Subcellular localization spans lysosomal membrane, late endosome membrane, cell membrane, and cytolytic granule membrane, undergoing dynamic trafficking between these compartments.
- Key post-translational modifications include extensive N-glycosylation and disulfide bond formation, contributing to its structural stability and function.
Experimental Guidance and Technical Tips
- The immunogen (aa 208–363) lies within the lumenal domain, suggesting the antibody recognizes an extracellular/lumenal epitope. For IF/ICC, permeabilization is required to access intracellular lysosomal LAMP1.
- For flow cytometry (FC) using this conjugate, direct staining of live cells may detect surface LAMP1 (e.g., on activated NK cells), while intracellular staining requires fixation and permeabilization for total LAMP1 detection.
- Consider co-staining with lysosomal markers (e.g., LAMP2, LysoTracker) and using knockout/knockdown controls to confirm specificity in immunofluorescence experiments.
