Biological Background: LAMP1 Function and Localization
- LAMP1 (Lysosome-associated membrane glycoprotein 1), also known as CD107a or LGP120, is a major protein component of the lysosomal membrane, heavily glycosylated with N- and O-linked glycans that form the protective glycocalyx lining the lysosomal lumen.
- It regulates lysosomal pH by acting as a direct inhibitor of the proton channel TMEM175, facilitating lysosomal acidification necessary for optimal hydrolase activity.
Related references: PMID:37390818 - LAMP1 is involved in autophagy and cholesterol homeostasis, playing a critical role in maintaining cellular catabolic balance.
- In NK cells, LAMP1 facilitates the movement of cytotoxic granules to the cell surface, mediates perforin trafficking to lytic granules, and protects NK cells from degranulation-associated damage by their own cytotoxic contents.
Related references: PMID:2022921, PMID:23632890, PMID:23847195 - Upon cell activation, LAMP1 can be expressed at the cell surface where it presents carbohydrate ligands to selectins, contributing to cell adhesion.
Related references: PMID:7685349 - LAMP1 acts as a receptor for Lassa virus glycoprotein and promotes viral fusion with host membranes in less acidic endosomes, highlighting its role in microbial infection.
Related references: PMID:24970085, PMID:25972533, PMID:27605678, PMID:29295909 - Subcellularly, LAMP1 is primarily found on lysosomal membranes, but also localizes to late endosomes, endosomes, the plasma membrane, and cytolytic granule membranes in immune cells.
- Post-translational modifications include extensive N- and O-linked glycosylation and disulfide bond formation, which are essential for its structural integrity and function.
Experimental Guidance and Technical Tips
- The immunogen for this antibody corresponds to a peptide region (aa 150-250) of human LAMP1. For Western blot, expect a band around 45 kDa, although highly variable glycosylation may cause the protein to migrate as a diffuse band at higher apparent molecular weights. Validate with appropriate positive controls.
- In immunofluorescence (IF/ICC), LAMP1 typically exhibits punctate perinuclear staining consistent with lysosomal localization. Use permeabilization and co-staining with organelle markers to confirm specificity.
- For immunohistochemistry (IHC-P), antigen retrieval and antibody titration are recommended to optimize signal-to-noise, particularly in tissues with high endogenous peroxidase activity.
- Cross-reactivity is confirmed for human, mouse, and rat, but performance in other species should be verified experimentally.
- This polyclonal antibody is suitable for ELISA; consider pairing with a monoclonal antibody for sandwich ELISA development if quantitative detection is required.
