Biological Background: LAMP1 Function and Localization
- LAMP1 (Lysosome-associated membrane glycoprotein 1) is a 45 kDa type I transmembrane glycoprotein and a member of the LAMP family, characterized by a heavily glycosylated luminal domain.
- It is essential for lysosome biogenesis and autophagy, and directly regulates lysosomal lumen pH by inhibiting the TMEM175 proton channel (PubMed:37390818). Related references: PMID:37390818
- LAMP1 participates in NK-cell-mediated cytotoxicity by facilitating cytotoxic granule movement and perforin trafficking to the lytic granule (PubMed:2022921, PubMed:23632890), and protects NK cells from self-inflicted damage during degranulation (PubMed:23847195). Related references: PMID:2022921 | PMID:23632890 | PMID:23847195
- The protein presents carbohydrate ligands to selectins, contributing to cell adhesion and migration (PubMed:7685349). Related references: PMID:7685349
- During Lassa virus infection, LAMP1 acts as a host receptor for the viral glycoprotein and promotes membrane fusion in less acidic endosomes (PubMed:24970085, PubMed:25972533, PubMed:27605678, PubMed:29295909). Related references: PMID:24970085 | PMID:25972533 | PMID:27605678 | PMID:29295909
- LAMP1 also assists the FURIN-mediated cleavage of mumps virus fusion protein F by interacting with both the protease and the unprocessed viral protein.
- Subcellularly, LAMP1 is predominantly found in lysosomal and late endosomal membranes, but can also localize to the cell surface and cytolytic granule membranes upon activation.
- The protein undergoes alternative splicing and contains multiple N-glycosylation sites and disulfide bonds essential for its stability and function.
Experimental Guidance and Technical Tips
- For Western blot, consider that LAMP1 is heavily glycosylated, which may cause it to migrate at a higher apparent molecular weight (~90–120 kDa) than the predicted 45 kDa. Use appropriate protein ladders and validate band specificity with control samples.
- Optimize blocking and wash conditions to reduce potential background, as polyclonal antibodies may exhibit higher non-specific binding.
- In ELISA, validate the antibody's binding specificity and optimal dilution using recombinant LAMP1 protein or cell lysates from human samples. Pair with a suitable detection system.
- This antibody is validated for human reactivity; cross-reactivity with other species is not expected based on provided data. Confirm suitability for other species if needed.
