Biological Background: Lamin A/C Function and Localization
- Lamin A and C are major components of the nuclear lamina, a filamentous meshwork underlying the inner nuclear membrane, and are encoded by the LMNA gene through alternative splicing.

- Lamins play a role in DNA repair: Lamin A/C is recruited to DNA double-strand breaks by XRCC4 and IFFO1, immobilizing broken DNA ends to prevent chromosome translocations. Related references: PMID:31548606

- The immunogen spans amino acids 403–572, corresponding to the C-terminal tail domain common to lamin A and C, which is less conserved among intermediate filament proteins, suggesting high specificity.
- For Western blot, the predicted molecular weight is ~74 kDa for lamin A and ~65 kDa for lamin C; both bands may be detected. Validate using nuclear extracts from human, mouse, or rat cell lines.
- For IHC-P and IF/ICC, nuclear envelope and nucleoplasmic staining is expected. Optimization of antigen retrieval and permeabilization may be required for formalin-fixed tissues.
- For IP and ELISA, the antibody may be used to enrich lamin A/C complexes; however, blocking and wash conditions should be empirically determined.
- Given the KO validation, this antibody is suitable for confirming lamin A/C depletion and studying loss-of-function phenotypes in nuclear organization and DNA repair assays.
