  • L3MBTL3 (Lethal(3)malignant brain tumor-like protein 3), also known as MBT-1, is encoded by the L3MBTL3 gene (KIAA1798, MBT1) in humans.
  • It functions as a negative regulator of Notch target gene expression, required for RBPJ-mediated transcriptional repression. Related references: PMID:29030483
  • L3MBTL3 recruits the histone demethylase KDM1A to Notch-responsive elements, promoting KDM1A-mediated H3K4me demethylation. Related references: PMID:29030483
  • It is involved in the regulation of ubiquitin-dependent degradation of methylated non-histone proteins, including SOX2, DNMT1, and E2F1, by acting as an adaptor that recruits the CRL4-DCAF5 E3 ubiquitin ligase complex to methylated target proteins.
