- KYNU (Kynureninase) is an enzyme also referred to as KYNUU or L-kynurenine hydrolase, encoded by the KYNU gene.
- It catalyzes the cleavage of L-kynurenine and L-3-hydroxykynurenine into anthranilic acid and 3-hydroxyanthranilic acid, respectively, playing a central role in tryptophan metabolism; it also has cysteine-conjugate-beta-lyase activity.
- Alternative splicing generates multiple isoforms; the canonical protein has a predicted molecular weight of ~52 kDa, while a 34 kDa isoform may also exist.
- Subcellular localization is cytoplasmic, primarily in the cytosol.
- KYNU is broadly expressed across human tissues, including heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas, with highest levels in placenta, liver, and lung; expression is also detected in all brain regions.
- The enzyme requires pyridoxal phosphate as a cofactor and is involved in pyridine nucleotide biosynthesis.
- Post-translational modifications include acetylation; it is part of the human reference proteome and disease-associated variants have been documented.
