Biological Background: KYAT3 Function and Localization
- Encodes kynurenine--oxoglutarate transaminase 3 (KYAT3), also known as kynurenine aminotransferase III (KATIII) and cysteine-S-conjugate beta-lyase 2 (CCBL2). Multiple isoforms are generated by alternative splicing.
- Catalyzes the irreversible transamination of L-kynurenine, an L-tryptophan metabolite, to form kynurenic acid (KA). KA acts as a broad-spectrum antagonist of ionotropic excitatory amino acid receptors, linking KYAT3 to neuromodulation and neuroprotection.
- Functions as a cysteine-S-conjugate beta-lyase, catalyzing the beta-elimination of S-conjugates and Se-conjugates of L-(seleno)cysteine, thereby cleaving C–S or C–Se bonds. This lyase activity suggests a role in the metabolism of xenobiotics and endogenous compounds.
- Exhibits broad substrate specificity in vitro, using glyoxylate as a preferred amino group acceptor. Active towards L-kynurenine, tryptophan, phenylalanine, serine, cysteine, methionine, histidine, glutamine, and asparagine. Lower activity is observed with 2-oxoglutarate.
- Belongs to the pyridoxal phosphate (PLP)-dependent aminotransferase family, consistent with its classification as a transferase. PLP serves as a critical cofactor for enzymatic activity.
