Biological Background: Ku80 Function and Localization
- The Ku80 protein, also known as XRCC5, KU86, KUB2, or NFIV, is the 86 kDa subunit of the Ku autoantigen, a heterodimeric DNA-binding complex essential for DNA double-strand break (DSB) repair via the classical non-homologous end joining (NHEJ) pathway. Together with XRCC6 (Ku70), it forms the Ku70:Ku80 heterodimer that directly binds DSB ends and serves as a scaffold for assembling the core NHEJ repair machinery, including PRKDC (DNA-PKcs), to align and ligate broken DNA ends.
