Biological Background: Ku80 Function and Localization

  • The Ku80 protein, also known as XRCC5, KU86, KUB2, or NFIV, is the 86 kDa subunit of the Ku autoantigen, a heterodimeric DNA-binding complex essential for DNA double-strand break (DSB) repair via the classical non-homologous end joining (NHEJ) pathway. Together with XRCC6 (Ku70), it forms the Ku70:Ku80 heterodimer that directly binds DSB ends and serves as a scaffold for assembling the core NHEJ repair machinery, including PRKDC (DNA-PKcs), to align and ligate broken DNA ends. Related references: PMID:11493912, PMID:33854234, PMID:34352203
  • Ku80 is indispensable for V(D)J recombination, a specialized NHEJ-driven process that generates the diverse repertoire of T-cell receptors and immunoglobulins during lymphocyte development. Related references: PMID:9742108
  • In addition to its structural role, Ku80 possesses 5'-deoxyribose-5-phosphate lyase activity, which cleans abasic sites near DSB termini to prepare them for ligation. A 3'-5' DNA helicase activity has also been reported, though its in vivo significance remains uncertain. Related references: PMID:20383123, PMID:7957065
  • Ku80 is implicated in telomere maintenance, contributing to the protection and regulation of chromosome ends. Related references: PMID:29490055
  • Beyond DNA repair, Ku80 functions as a transcriptional cofactor, regulating gene expression by interacting with various transcription factors. Related references: PMID:12145306, PMID:8621488
  • Ku80 participates in the innate immune response to DNA viruses by assembling into the HDP-RNP complex, which acts as a platform for IRF3 phosphorylation and activates the cGAS-STING pathway. Related references: PMID:28712728
  • Ku80 can bind cellular RNAs and recruit PRKDC, playing a role in ribosome biogenesis, particularly through interaction with U3 small nucleolar RNA. Related references: PMID:32103174
  • Localized predominantly in the nucleus and nucleolus, Ku80 associates with chromatin and is subject to multiple post-translational modifications, including acetylation, phosphorylation, ADP-ribosylation, and ubiquitin-like conjugation.

Experimental Guidance and Technical Tips

  • Immunogen design: The antibody was raised against a synthetic peptide spanning amino acids 633–732 of human Ku80. When designing validation experiments, consider that this region may be subject to post-translational modifications or alternative splicing events that could affect antibody binding.
  • Western blotting (WB): Expect a specific band at approximately 83 kDa in human cell lysates. Include appropriate positive and negative controls (e.g., Ku80 knockout or siRNA-treated samples) to confirm specificity.
  • Immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC): Ku80 exhibits nuclear and nucleolar localization. Optimize fixation and permeabilization conditions for your tissue or cell type, and validate staining with a known nuclear marker.
  • Immunoprecipitation (IP): This antibody can be used to pull down the Ku70/Ku80 complex. Co-immunoprecipitation of Ku70 (XRCC6) serves as a functional control.
  • ELISA: Suitable for direct or sandwich ELISA formats. Validate cross-reactivity with other species if needed, as the antibody is specified for human reactivity.
