Biological Background: Ku80 Function and Localization
- Encoded by XRCC5, Ku80 (also known as Ku86, NFIV, and lupus autoantigen p86) localizes to the nucleus, nucleolus, and chromosomes, forming a stable heterodimer with Ku70 (XRCC6).
- The Ku complex is the primary sensor for DNA double-strand breaks (DSBs), initiating canonical non-homologous end joining (NHEJ) and V(D)J recombination. This pathway ensures rapid repair of cytotoxic DSBs and contributes to immune diversity. Related references: PMID:11493912, PMID:33854234, PMID:34352203, PMID:9742108
- Likely possesses 5'-deoxyribose-5-phosphate lyase (5'-dRP lyase) activity, catalyzing beta-elimination at abasic sites near DSBs to clean broken ends prior to ligation. Related references: PMID:20383123
- May also have 3'-5' DNA helicase activity, though its physiological relevance in vivo remains unconfirmed. Related references: PMID:7957065
- Plays a role in telomere maintenance, contributing to genomic stability. Related references: PMID:29490055
- Acts as a transcriptional cofactor for various transcription factors, thereby regulating gene expression. Related references: PMID:12145306, PMID:8621488
- Involved in innate immunity by assembling into the HDP-RNP complex, which serves as a platform for IRF3 phosphorylation and activation of the cGAS-STING pathway. Related references: PMID:28712728
- Binds RNAs and recruits PRKDC to a wide range of cellular RNAs, including U3 small nucleolar RNA, thereby playing a role in ribosomal RNA biogenesis. Related references: PMID:32103174
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (amino acids 543–732) of human Ku80, a domain conserved across species; this may account for the observed cross-reactivity with mouse and rat.
- For Western blotting, the predicted molecular weight is 83kDa; consider validating the band in nuclear-enriched lysates from human, mouse, or rat cell lines.
- The antibody is reactive in multiple applications (WB, IHC-P, IF/ICC, IP, ELISA); optimize dilutions and incubation conditions for each assay.
- In immunofluorescence, Ku80’s nuclear and nucleolar localization suggests that permeabilization with detergents (e.g., Triton X-100) may be necessary to achieve optimal staining.
