Biological Background: Ku70 Function and Localization
- Ku70 (gene XRCC6) is a DNA-binding protein critical for the repair of DNA double-strand breaks (DSBs) via the classical non-homologous end joining (NHEJ) pathway. It forms the Ku70:Ku80 heterodimer that binds DSB ends and recruits core NHEJ factors to facilitate broken end alignment and ligation. Related references: PMID:11493912, PMID:20493174, PMID:33854234
- Ku70 also functions as a 5'-deoxyribose-5-phosphate lyase (5'-dRP lyase), removing abasic sites near breaks to clean termini for ligation. Related references: PMID:20383123
- The Ku complex plays a role in V(D)J recombination, contributing to immune diversity by processing programmed DSBs. Related references: PMID:9742108
- Ku70 is involved in telomere maintenance and telomere capping. Related references: PMID:29490055
- It acts as a transcriptional cofactor, interacting with various transcription factors to regulate gene expression. Related references: PMID:12145306, PMID:8621488
- In innate immunity, Ku70 participates in the HDP-RNP complex that activates IRF3 and the cGAS-STING pathway in response to DNA viruses. Related references: PMID:28712728
- Ku70 negatively regulates apoptosis by binding to BAX and sequestering it from mitochondria, and may deubiquitinate BAX. Related references: PMID:15023334, PMID:18362350, PMID:35545041
- Ku70 is predominantly nuclear but also localizes to chromosomes and the cytoplasm, and undergoes post-translational modifications including acetylation, phosphorylation, and ADP-ribosylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 500–609 of human Ku70 (P12956). This region may be conserved across species; cross-reactivity with mouse and rat is indicated. Validate reactivity in your target sample system.
- For Western blot (WB), the predicted molecular weight is approximately 70 kDa. Optimize blocking and antibody dilution to minimize nonspecific binding.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), appropriate antigen retrieval and permeabilization methods should be tested, as Ku70 localizes mainly to the nucleus.
- ELISA application is supported; pairing with a suitable detection antibody is recommended for sandwich ELISA development.
