Biological Background: KTN1 Function and Localization

  • KTN1 encodes the protein kinectin, also known as CG-1 antigen or kinesin receptor, which acts as a receptor for kinesin, facilitating kinesin-driven vesicle motility along microtubules.
  • Upon fibronectin-induced integrin aggregation, kinectin accumulates in integrin-based adhesion complexes (IAC), linking vesicle transport to cell adhesion.
  • Kinectin is a single-pass type I membrane protein anchored in the endoplasmic reticulum (ER) membrane, with its functional N-terminus facing the cytoplasm.
  • The protein undergoes alternative splicing and contains coiled-coil regions, suggesting roles in protein–protein interactions.
  • Post-translational modifications include phosphorylation and glycosylation, which may regulate its activity and localization.
  • Tissue expression is highest in peripheral blood lymphocytes, testis, and ovary, with lower levels in spleen, thymus, prostate, small intestine, and colon.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from the N-terminal region (aa 1–100) of human KTN1, which is conserved across human, mouse, and rat, supporting the observed cross-reactivity. When performing Western blot, consider using lysates from testis or ovary, where expression is high, to detect the ~156 kDa band.
  • For ELISA applications, the antibody may be used as a capture or detection reagent; optimize working concentrations with your specific antigen and assay format.
  • Given the ER membrane localization, subcellular fractionation or enrichment steps may improve detection sensitivity in low-expressing samples.
  • As a polyclonal antibody, lot-to-lot consistency should be verified, and working dilutions should be titrated for each new batch.
