Biological Background: KSR1 Function and Localization
- KSR1 (Kinase suppressor of Ras 1) is a scaffold protein and putative serine/threonine-protein kinase that plays a central role in the MAPK signaling cascade.
- It forms part of a multiprotein signaling complex that promotes phosphorylation of Raf family members and activation of downstream MAP kinases.
- Acts as a MAP2K1/MEK1- and MAP2K2/MEK2-dependent allosteric activator of BRAF; upon binding to MEK, it dimerizes with BRAF and stimulates BRAF-mediated MEK phosphorylation (PubMed:29433126). Related references: PMID:29433126
- Promotes activation of MAPK1/ERK2 and MAPK3/ERK1 in response to EGF and cAMP stimulation.
- Subcellular localization includes cytoplasm, cell membrane, ruffle membrane, and endoplasmic reticulum membrane, consistent with its dynamic signal transduction roles.
- Contains a zinc-finger domain and exhibits ATP-binding activity; however, its intrinsic kinase activity remains disputed and may be context-dependent.

Experimental Guidance and Technical Tips
- The immunogen (aa 320–420) lies outside the kinase domain; sequence homology with mouse and rat KSR1 supports cross-reactivity, but alignment should be verified for the exact epitope region.
- For Western blot, a band near 102 kDa is expected; optimize lysis and loading conditions using positive control cell lines (e.g., HeLa, NIH/3T3).
- In immunofluorescence/ICC, test both methanol and formaldehyde fixation, as KSR1 localizes to membrane ruffles and the endoplasmic reticulum; permeabilization with Triton X-100 or digitonin may be necessary.
- For ELISA, consider pairing with a detection antibody against a non-overlapping epitope after validating binding affinity and specificity.
