Biological Background: KRT81 Function and Localization
- KRT81, also known as Keratin 81 (aliases: HB1, K81, KRTHB1, MLN137), is a type II hair keratin that belongs to the intermediate filament family, characterized by coiled-coil domains and potential for isopeptide bond and ubiquitin-like conjugation modifications.
- As a structural component, KRT81 forms obligate heterodimers with type I hair keratins, which assemble into keratin intermediate filaments essential for the mechanical strength and integrity of the hair shaft. Related references: PMID:10504448, PMID:9402962
- It is abundantly expressed in the differentiating cortex of growing (anagen) hair follicles, specifically in keratinocytes of the hair cortex, and is absent from the inner root sheath and medulla. Related references: PMID:11445569
- KRT81 is also detected in malignant lymph node tissue associated with breast carcinoma, and Uniprot keywords associate it with disease variants, suggesting roles beyond hair biology.

- The immunogen is a synthetic peptide corresponding to amino acids 350–450 of human KRT81 (NP_002272.2). This C-terminal region was selected to potentially avoid cross-reactivity with other keratins; however, sequence homology should be verified against related type II keratins.
- For Western blotting, expect a single band at approximately 55kDa in human hair follicle lysates or appropriate tissue extracts; using reducing conditions may aid in solubilizing keratin filaments. Validate with mouse samples as cross-reactivity is indicated; cross-reactivity with other species should be determined empirically.
- In ELISA, the antibody can be employed for direct antigen detection; consider peptide competition assays to confirm specificity if needed.
- Due to the insolubility of keratins, include a denaturing and reducing sample buffer during lysis and consider sonication to release KRT81 from the intermediate filament network.
