  • KRT81, also known as Keratin 81 (aliases: HB1, K81, KRTHB1, MLN137), is a type II hair keratin that belongs to the intermediate filament family, characterized by coiled-coil domains and potential for isopeptide bond and ubiquitin-like conjugation modifications.

  • It is abundantly expressed in the differentiating cortex of growing (anagen) hair follicles, specifically in keratinocytes of the hair cortex, and is absent from the inner root sheath and medulla. Related references: PMID:11445569

  • For Western blotting, expect a single band at approximately 55kDa in human hair follicle lysates or appropriate tissue extracts; using reducing conditions may aid in solubilizing keratin filaments. Validate with mouse samples as cross-reactivity is indicated; cross-reactivity with other species should be determined empirically.
  • In ELISA, the antibody can be employed for direct antigen detection; consider peptide competition assays to confirm specificity if needed.
  • Due to the insolubility of keratins, include a denaturing and reducing sample buffer during lysis and consider sonication to release KRT81 from the intermediate filament network.
