Biological Background: KRT10 Function and Localization
- Keratin 10 (K10/CK-10) is a type I cytoskeletal keratin that forms intermediate filaments, providing mechanical support to suprabasal keratinocytes.
- Obligate heteropolymer partner for type II keratins KRT1 and KRT2, critical for terminal differentiation and epidermal barrier function.
- Acts as a receptor for Staphylococcus aureus clfB on nasal epithelial cells, facilitating nasal colonization.
- Mediates Streptococcus pneumoniae adherence to lung epithelia via PsrP binding, influencing bacterial infection.
- Localized intracellularly in the cytoplasm, on the cell surface, and secreted extracellularly.
- Expressed throughout suprabasal epidermal layers (including stratum corneum) and on the surface of lung cell lines and desquamated nasal epithelial cells.

Experimental Guidance and Technical Tips
- This rabbit polyclonal antibody targets a synthetic peptide within amino acids 350–460 of human KRT10 (NP_000412.3). For Western blot, expect a band near 59 kDa; use appropriate positive controls such as human keratinocyte lysates.
- For ELISA, optimize coating conditions and consider using the immunogen peptide as a positive control for direct or competitive ELISA formats.
- Although reported to be human-specific, cross-reactivity with KRT10 from other species has not been tested; validate in the relevant sample system.
- Polyclonal antibodies may exhibit lot-to-lot variability; confirm performance with reference standards and include negative controls.
