Biological Background: KREMEN1 Function and Localization

  • KREMEN1 (Kremen protein 1), also known as Dickkopf receptor, kringle domain-containing transmembrane protein 1, or kringle-containing protein marking the eye and the nose, is a single-pass type I transmembrane receptor.
  • It functions as a receptor for Dickkopf (DKK) proteins, cooperating with DKK1/2 to inhibit Wnt/β-catenin signaling by promoting the endocytosis of LRP5/6 co-receptors.
  • In the absence of DKK ligands, KREMEN1 can potentiate Wnt/β-catenin signaling by maintaining LRP5 or LRP6 at the cell membrane.
  • KREMEN1 plays critical roles in development: it attenuates Wnt signaling in the developing limb to ensure normal limb patterning, and negatively regulates bone formation.
  • The protein contains a kringle domain and undergoes post-translational modifications including glycosylation and disulfide bond formation.
  • KREMEN1 is localized to the cell membrane.
  • Alternative splicing produces multiple isoforms.
  • It has a Wnt-independent apoptotic activity, which is inhibited by DKK1 binding.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 180-380 of human KREMEN1 (NP_001034659.2), which lies within the extracellular region, likely targeting the kringle domain. Consider verifying reactivity with relevant isoforms in your sample system.
  • For Western blot, a band around 52 kDa is expected; optimize loading and antibody dilution for your specific cell or tissue lysates.
  • This polyclonal antibody is suitable for ELISA; consider pairing with a matched detection antibody for sandwich ELISA development.
