Biological Background: KPNA6 Function and Localization

  • Importin subunit alpha-7 (KPNA6) is a member of the importin alpha family and functions as an adapter for the nuclear transport receptor karyopherin subunit beta-1 (KPNB1).
  • It specifically recognizes and binds to cargo proteins containing either a simple or bipartite nuclear localization signal (NLS).
  • The importin alpha/beta heterodimer docks at the nuclear pore complex (NPC) via KPNB1 binding to FG-repeat nucleoporins, and translocation through the NPC is driven by a RanGTP-dependent mechanism.
  • Directional nuclear import is conferred by the asymmetric distribution of RanGTP (high in the nucleus) and RanGDP (high in the cytoplasm), leading to cargo release in the nucleus.
  • After cargo release, importin alpha and beta are recycled back to the cytoplasm for another round of transport.
  • KPNA6 is widely expressed across tissues.
  • Post-translational modifications include acetylation and phosphorylation; the protein is also implicated in host-virus interactions.
  • The predicted molecular weight is approximately 60 kDa (60030 Da).

  • The immunogen is a synthetic peptide spanning amino acids 69–169 of human KPNA6; consider performing a peptide competition or knockdown experiment to confirm band specificity in your system.
  • For Western blot, a starting dilution of 1:1000–1:2000 may be used, but optimal concentration should be determined empirically.
  • For immunofluorescence (IF/ICC), include appropriate fixation/permeabilization steps; test both methanol and paraformaldehyde protocols.
  • Cross-reactivity with human and mouse is specified; when using mouse samples, verify antibody specificity as endogenous IgG may interfere if using anti-mouse secondary.
