- It binds specifically to nuclear localization signals (NLS), either simple or bipartite, on cargo proteins and links them to importin-beta (KPNB1) for translocation through the nuclear pore complex.
- The nuclear import cycle is regulated by Ran GTPase, with directionality conferred by the asymmetric distribution of GTP- and GDP-bound Ran across the nuclear envelope.
- It is localized in both the cytoplasm and the nucleus, shuttling between these compartments as part of the importin recycling process.
- In vitro, it mediates the nuclear import of human cytomegalovirus UL84 via a non-classical NLS and recognizes the NLS of influenza A virus nucleoprotein, highlighting its role in host-virus interactions.
- It is ubiquitously expressed, with highest levels in heart and skeletal muscle, and is subject to acetylation and phosphorylation.
