Biological Background: KPNA1 Function and Localization

  • KPNA1, also known as Importin subunit alpha-5, Karyopherin subunit alpha-1, NPI-1, RCH2, and SRP1-beta, is a member of the importin alpha family that functions as an adapter for nuclear protein import.
  • Functions as an adapter for nuclear protein import by specifically binding to substrates containing simple or bipartite NLS motifs, and docking to the nuclear pore complex via interaction with KPNB1, followed by Ran-dependent translocation.   • Mediates nuclear import of the PR-DUB complex component BAP1, acting redundantly with KPNA2 and Transportin-1/TNPO1. Related references: PMID:35446349
  • In vitro, mediates the nuclear import of human cytomegalovirus UL84 protein by recognizing a non-classical NLS, implicating KPNA1 in host-virus interactions.
  • Localizes to both the cytoplasm and nucleus, consistent with its role in shuttling cargoes across the nuclear envelope.
  • Expressed ubiquitously, indicating a fundamental housekeeping role in nuclear transport across tissues.
  • Subject to acetylation and phosphorylation, and is a target for ubiquitin-like (Ubl) conjugation, as identified by proteomics studies; these modifications may regulate its activity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-300) of human KPNA1, which contains the NLS-binding domain; consider using appropriate controls to confirm specificity in your system.
  • Validated for Western blot (WB) and ELISA applications. For WB, the predicted molecular weight is approximately 60 kDa; validate with positive control lysates from human or mouse cells.
  • Cross-reacts with human and mouse KPNA1. When working with other species, check sequence homology before use.
  • For ELISA, perform a titration to determine optimal working concentration under your assay conditions.
