Biological Background: Calpain-2 Function and Localization
- Calpain-2, also known as M-calpain, is the large catalytic subunit of the calcium-activated neutral protease calpain-2, encoded by the CAPN2 gene. It belongs to the peptidase C2 family and functions as a heterodimer with a small regulatory subunit.
- As a calcium-regulated non-lysosomal thiol-protease, it catalyzes limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction, playing a role in cell migration, proliferation, and differentiation.
- Calpain-2 proteolytically cleaves myocilin (MYOC) at Arg-226, a cleavage event implicated in the regulation of intraocular pressure (PubMed:17650508). Related references: PMID:17650508
- Following neuronal stimulation, calpain-2 cleaves CPEB3, which abolishes its translational repressor activity, thereby enabling translation of CPEB3 target mRNAs (By similarity).

Experimental Guidance and Technical Tips
- This antibody was generated using a recombinant fusion protein spanning amino acids 350–680 of human Calpain-2 (NP_001739.2), a region covering part of the catalytic domain and the C2-like calcium-binding domain. Consider this immunogen design when validating reactivity in your model system.
- For Western blot (WB) applications, expect a band at the predicted molecular weight of ~80 kDa. Note that calpain-2 may undergo autocatalytic processing, which could result in additional lower molecular weight fragments.
- The antibody is also suitable for immunohistochemistry (IHC-P) and ELISA. When performing IHC, include appropriate controls and optimize antigen retrieval conditions for the tissues of interest.
- Cross-reactivity has been observed with human, mouse, and rat samples. For other species, verify sequence homology, particularly in the immunogen region, before use.
- Since calpain-2 activity is calcium-dependent, consider the calcium concentration in your lysis buffers and experimental conditions, as it may affect the protease's autolysis and epitope availability.
