Biological Background: KMT2B Function and Localization
- KMT2B (Histone-lysine N-methyltransferase 2B), also known as MLL4, TRX2, and WBP7, is a large (~294 kDa) chromatin-modifying enzyme belonging to the mixed-lineage leukemia (MLL) family of histone methyltransferases.
- It catalyzes the transfer of methyl groups from S-adenosyl-L-methionine to lysine 4 of histone H3 (H3K4), predominantly forming monomethyl (H3K4me1) and dimethyl (H3K4me2) marks via a non-processive mechanism, thereby regulating gene transcription and DNA repair (PubMed:17707229, PubMed:25561738). - KMT2B plays a redundant role with KMT2C in enriching H3K4me1 on primed and active enhancer elements, contributing to cell type-specific gene expression programs (PubMed:24081332). Related references: PMID:24081332
- It localizes to the nucleus and is recruited by transcription factors such as NFE2 to control beta-globin locus transcription. It is essential for establishing/maintaining H3K4me3 during oocyte growth and preimplantation development.
- KMT2B is widely expressed, with highest levels in testis and elevated expression in cerebellum, and is frequently amplified in pancreatic carcinomas. Mutations are linked to dystonia and intellectual disability.
- The protein contains multiple conserved domains, including a SET domain for methyltransferase activity, PHD zinc fingers, a bromodomain, and DNA-binding motifs, highlighting its multifunctional role in chromatin regulation.

- For Western blotting, use appropriate nuclear extracts or whole-cell lysates from human, mouse, or rat cells/tissues. Expect a band around 294 kDa; optimize transfer conditions (e.g., low methanol, extended transfer time) for large proteins.
