Biological Background: KMT2A Function and Localization

  • Histone-lysine N-methyltransferase 2A (KMT2A), also known as MLL, HRX, or ALL-1, is a member of the Trithorax family of chromatin regulators. It plays an essential role in early development and hematopoiesis through histone H3K4 methylation (PubMed:12453419, PubMed:15960975, PubMed:19187761). Related references: PMID:12453419 PMID:15960975 PMID:19187761
  • KMT2A is the catalytic subunit of the MLL1/MLL complex, which mediates both H3K4 methylation and H4K16 acetylation, thereby regulating chromatin accessibility at active gene promoters.
  • It binds unmethylated CpG elements in promoter regions and helps maintain their nonmethylated state. This activity is required for transcriptional activation of target genes such as HOXA9 (PubMed:20010842, PubMed:20677832). Related references: PMID:20010842 PMID:20677832
  • KMT2A promotes PPP1R15A-induced apoptosis, highlighting a role in programmed cell death (PubMed:10490642). Related references: PMID:10490642
  • In the circadian clock, KMT2A establishes a permissive chromatin state through rhythmic H3K4 methylation, which facilitates CLOCK-BMAL1 recruitment and circadian gene expression (By similarity).
  • The protein exhibits auto-methylation activity on cysteine 3882 in the absence of histone substrates (PubMed:24235145). Related references: PMID:24235145
  • KMT2A is predominantly localized in the nucleus and is expressed in heart, lung, brain, and T- and B-lymphocytes. It is classified as a proto-oncogene and is frequently involved in chromosomal rearrangements associated with acute leukemias.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 752–949 of human KMT2A, located in the N-terminal region. This antibody may detect full-length KMT2A as well as N-terminal fragments or fusion proteins derived from chromosomal translocations, making it potentially useful for studying MLL-rearranged leukemias.
  • Due to the large molecular weight of KMT2A (~432 kDa), consider using low-percentage SDS-PAGE gels (e.g., 6–8%) and extended wet transfer times (>4 hours at 30 V) for western blotting. Coomassie staining of the gel after transfer can help confirm efficient protein migration.
  • For western blot, validate the antibody in knockout (KO) cell lines or tissue samples to confirm specificity, as indicated by the KO validation status. Include appropriate positive and negative controls.
  • In ELISA applications, the polyclonal nature of the antibody provides broad epitope coverage but may require careful blocking and washing optimization to reduce background.
