Biological Background: KMT2A Function and Localization
- Histone-lysine N-methyltransferase 2A (KMT2A), also known as MLL, HRX, or ALL-1, is a member of the Trithorax family of chromatin regulators. It plays an essential role in early development and hematopoiesis through histone H3K4 methylation (PubMed:12453419, PubMed:15960975, PubMed:19187761). - KMT2A is the catalytic subunit of the MLL1/MLL complex, which mediates both H3K4 methylation and H4K16 acetylation, thereby regulating chromatin accessibility at active gene promoters.

- The immunogen corresponds to amino acids 752–949 of human KMT2A, located in the N-terminal region. This antibody may detect full-length KMT2A as well as N-terminal fragments or fusion proteins derived from chromosomal translocations, making it potentially useful for studying MLL-rearranged leukemias.
- Due to the large molecular weight of KMT2A (~432 kDa), consider using low-percentage SDS-PAGE gels (e.g., 6–8%) and extended wet transfer times (>4 hours at 30 V) for western blotting. Coomassie staining of the gel after transfer can help confirm efficient protein migration.
- For western blot, validate the antibody in knockout (KO) cell lines or tissue samples to confirm specificity, as indicated by the KO validation status. Include appropriate positive and negative controls.
- In ELISA applications, the polyclonal nature of the antibody provides broad epitope coverage but may require careful blocking and washing optimization to reduce background.
