Biological Background: KLRD1 Function and Localization
- KLRD1 (CD94) is a type II integral membrane glycoprotein belonging to the killer cell lectin-like receptor family. It functions as a ligand-binding subunit that pairs with KLRC1 or KLRC2 to form functional heterodimers.
- The KLRD1-containing complex recognizes the non-classical MHC class Ib molecule HLA-E loaded with self-peptides, enabling immune cells to monitor MHC class I expression and maintain self-tolerance. Related references: PMID:10023772, PMID:18064301, PMID:18083576
- The KLRD1-KLRC1 complex transmits inhibitory signals through ITIM-mediated recruitment of SHIP-1 and SHIP-2 phosphatases, counteracting activating receptor signals. This inhibitory function regulates NK and T cell activation, and in tumor microenvironments may contribute to immune cell exhaustion. Related references: PMID:30860984, PMID:9430220, PMID:9485206, PMID:12387742, PMID:18064301, PMID:30503213, PMID:12165520
- The KLRD1-KLRC2 complex acts as an activating receptor, signaling through the DAP12 adapter protein upon HLA-E binding. This pathway is important for adaptive NK cell responses and antiviral immunity. Related references: PMID:15940674, PMID:9655483, PMID:30134159, PMID:9754572, PMID:20952657, PMID:21825173
- KLRD1 is primarily located on the cell membrane and is expressed in NK cells, subsets of CD8-positive T cells, and intraepithelial lymphocytes. Its expression is differentially regulated in various lymphocyte populations. Related references: PMID:21825173, PMID:9430220, PMID:9485206, PMID:12387742, PMID:20952657
- The protein undergoes post-translational modifications, including disulfide bond formation and glycosylation.
- Viruses such as human cytomegalovirus and HIV-1 exploit HLA-E/KLRD1 interactions to evade NK cell cytotoxicity. SARS-CoV-2 may also induce NK cell exhaustion through this pathway. Related references: PMID:32859121
Experimental Guidance and Technical Tips
- The immunogen (amino acids 32-179) encompasses the extracellular C-type lectin domain, which is predicted to be accessible for antibody binding.
- For Western blot analysis, the predicted molecular weight is 21 kDa; however, glycosylation may result in a higher apparent molecular weight.
- Validate the antibody in positive control lysates from NK cell lines (e.g., NK-92) or primary human NK cells. Cross-reactivity with mouse allows testing in mouse splenocyte or NK cell lysates.
- For ELISA applications, the antibody can be used for antigen capture or detection following standard indirect ELISA protocols. Optimization of coating and detection conditions is recommended.
