Biological Background: KLRC4 Function and Localization
- KLRC4 encodes NKG2-F, a type II integral membrane protein also known as NKG2F or NKG2-F-activating NK receptor.
- It belongs to the NKG2 family of C-type lectin-like receptors expressed primarily on natural killer (NK) cells.
- NKG2-F may function as a receptor for MHC class I HLA-E molecules, contributing to NK cell recognition and immune surveillance.
- The protein contains a signal-anchor transmembrane domain and is localized to the cell membrane.
- Post-translational modifications include at least one disulfide bond, consistent with its structure as a C-type lectin-like receptor.
- No tissue expression outside NK cells has been reported, indicating a specialized role in innate immunity.
Experimental Guidance and Technical Tips
- For Western blot, consider using this polyclonal antibody to detect the ~18 kDa KLRC4 protein in NK cell lysates; optimize loading and detection conditions.
- When performing immunofluorescence (IF/ICC or IF-P), validate fixation and permeabilization protocols, as the target is a transmembrane protein.
- The antibody cross-reacts with mouse and rat KLRC4; verify specificity in these species using appropriate positive and negative controls.
- For ELISA applications, titrate the antibody and use a suitable coating antigen to establish a sensitive detection system.
- Given that the immunogen spans the N-terminal region (aa 1–55), the antibody likely recognizes epitopes within the cytoplasmic domain; consider this for epitope mapping experiments.
