Biological Background: NKG2-A/NKG2-B Function and Localization
- Protein identity and isoforms: KLRC1 encodes NKG2-A and NKG2-B isoforms, which are type II transmembrane receptors of the NKG2 family.
- Immune inhibitory receptor function: NKG2-A/NKG2-B form heterodimers with KLRD1 and recognize HLA-E loaded with self-peptides, enabling cells to monitor MHC class I expression and maintain self-tolerance. Related references: PMID:18083576, PMID:37264229, PMID:9430220
- Signaling mechanism: Upon ligand binding, ITIMs in the intracellular domain recruit phosphatases SHP-1 and SHP-2, which dephosphorylate proximal signaling molecules to dampen activating signals. Related references: PMID:12165520, PMID:9485206
- Role in NK cells: NKG2-A is a dominant inhibitory receptor on NK cells that regulates their activation and prevents cytotoxicity against healthy cells. Related references: PMID:30860984, PMID:9430220, PMID:9485206
- Regulation of T cell responses: On CD8+ T cells, it counters TCR signaling to avoid autoimmunity; on intraepithelial γδ T cells, it triggers TGF-β1 secretion to limit cytotoxic αβ T cell responses. Related references: PMID:12387742, PMID:18064301
- Tumor immune checkpoint: In the tumor microenvironment, NKG2-A engagement by HLA-E can drive NK and T cell exhaustion, contributing to immune evasion. Related references: PMID:30503213, PMID:30860984
- Subcellular localization: Localizes to the cell membrane as a type II integral membrane protein.
- Tissue distribution: Predominantly expressed in NK cells and subsets of CD8+ T cells, including intraepithelial and intratumoral populations.
Experimental Guidance and Technical Tips
- The immunogen (aa 96–215) encompasses the extracellular C-type lectin-like domain of NKG2-A, which mediates HLA-E binding; this region is likely to generate antibodies that recognize native conformations.
- For western blot, a band around 26 kDa is expected; use denaturing and reducing conditions. Validate in relevant human or mouse lymphocyte samples, as the antibody cross-reacts with mouse.
- For ELISA, this polyclonal antibody can be used for detection; titrate the antibody to achieve the desired signal-to-noise ratio and include appropriate controls.
- Cross-reactivity with mouse suggests conservation of the epitope; verify cross-reactivity in your specific experimental setup.
- Since NKG2-A expression can be regulated under various conditions, consider including appropriate positive and negative controls (e.g., NK cell lines or activated T cells).
