Biological Background: KLKB1 Function and Localization
- KLKB1 encodes plasma prekallikrein, the zymogen of plasma kallikrein, a serine protease that circulates in blood as an inactive precursor.
- Upon activation, plasma kallikrein participates in the surface-dependent activation of blood coagulation by reciprocally activating coagulation factor XII (FXII) after binding to negatively charged surfaces.
- It releases the vasoactive peptide bradykinin from high-molecular-weight kininogen (HMWK), linking the contact activation system to inflammation and vascular permeability.
- Plasma kallikrein may also contribute to the renin-angiotensin system by converting prorenin into active renin.
- Subcellular localization is exclusively extracellular; the protein is secreted into plasma, where it is normally found.
- The protein contains a signal peptide, multiple Apple domains for protein–protein interactions, and a trypsin-like serine protease domain; it is N-glycosylated and stabilized by disulfide bonds.
- Genetic variants in KLKB1 are associated with prekallikrein deficiency (Fletcher trait), a rare condition with prolonged activated partial thromboplastin time but typically no bleeding tendency.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 20–300 of the heavy chain, which contains the four Apple domains responsible for binding to HMWK and negatively charged surfaces; this region is highly conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blotting, a band near 71 kDa (prekallikrein) or ~85 kDa (full-length with glycosylation) may be detected; reduction with DTT/TCEP is recommended to resolve disulfide-linked chains.
- In immunohistochemistry (IHC-P), consider antigen retrieval with citrate buffer (pH 6.0) and a detection system sensitive for rabbit primary antibodies; pre-immune serum or blocking peptide controls are advised to confirm specificity.
- For ELISA, the antibody can be used as capture or detection reagent; however, pairing with a monoclonal antibody to a distinct epitope may improve sensitivity and specificity.
- Because plasma kallikrein is abundant in blood, pre-adsorption of the polyclonal antibody against immobilized mouse/rat serum proteins can reduce background in rodent tissue applications if non-specific binding is observed.
