Biological Background: Kallikrein-15 Function and Localization
- Kallikrein-15 (KLK15), also known as ACO protease, is a member of the tissue kallikrein family of serine proteases.
- The physiological substrate of KLK15 remains unknown; it is classified as a hydrolase with predicted serine protease activity.
- KLK15 is encoded by the KLK15 gene and is expressed as a secreted zymogen that likely requires proteolytic processing for activation.
- Highest expression levels are found in the thyroid gland, with detectable expression also in the prostate, salivary glands, adrenal glands, colon, testis, and kidney.
- The protein contains disulfide bonds and undergoes glycosylation, features common to secreted proteases.
- Alternative splicing generates multiple isoforms, and the reference proteome entry lists a molecular weight of approximately 28 kDa.
- UniProtKB keywords include: Alternative splicing, Disulfide bond, Glycoprotein, Hydrolase, Protease, Secreted, Serine protease, and Zymogen.
Experimental Guidance and Technical Tips
- The immunogen, a recombinant fragment corresponding to amino acids 80-180, was designed to target a region unique to human KLK15; consider this when assessing detection specificity.
- For Western blot, validate antibody performance under reducing conditions using lysates from tissues with known KLK15 expression, such as thyroid or prostate.
- Cross-reactivity with mouse and rat orthologs is reported; verify and optimize for each species using appropriate positive and negative controls.
- ELISA conditions (coating antigen concentration, antibody dilution, and blocking) should be empirically determined, as no standardized protocol is supplied.
