Biological Background: Kallikrein-1 Function and Localization
- KLK1 encodes kallikrein-1, also known as tissue kallikrein, a secreted serine protease of the glandular kallikrein family.
- The primary function of KLK1 is the cleavage of kininogen to release Lys-bradykinin, a potent vasoactive peptide involved in blood pressure regulation and inflammation.
- In the context of microbial infection, KLK1 cleaves the Neisseria meningitidis NHBA protein in saliva, suggesting a role in innate immune defense at mucosal surfaces.
- Alternative splicing yields multiple isoforms; isoform 2 is detected in pancreas, salivary glands, kidney, colon, prostate, testis, and spleen.
- The zymogen form (prokallikrein) is activated by proteolytic removal of the propeptide, and the mature enzyme contains conserved disulfide bonds and N-linked glycosylation sites.
- As a member of the tissue kallikrein family, KLK1 shares structural homology with other kallikreins implicated in physiological and pathological processes, including cancer.
Experimental Guidance and Technical Tips
- The immunogen (aa 25–262) spans the mature KLK1 protein; thus the antibody may recognize both the propeptide and active forms. Validate band patterns using appropriate controls.
- For Western blot, a primary band near 29 kDa is anticipated, but glycosylation can cause minor mobility shifts. Use reducing conditions.
- When performing IHC-P, including an antigen retrieval step is recommended to expose epitopes in formalin-fixed samples. Testing dilution series is advised.
- The antibody shows cross-reactivity with mouse and rat KLK1 by supplier testing; however, confirm reactivity in your specific tissue or cell line to ensure specificity.
- For ELISA, the polyclonal nature makes it suitable as both capture and detection reagent; optimize pairing conditions to minimize background.
