  • The primary function of KLK1 is the cleavage of kininogen to release Lys-bradykinin, a potent vasoactive peptide involved in blood pressure regulation and inflammation.
  • In the context of microbial infection, KLK1 cleaves the Neisseria meningitidis NHBA protein in saliva, suggesting a role in innate immune defense at mucosal surfaces.
  • Alternative splicing yields multiple isoforms; isoform 2 is detected in pancreas, salivary glands, kidney, colon, prostate, testis, and spleen.
  • The zymogen form (prokallikrein) is activated by proteolytic removal of the propeptide, and the mature enzyme contains conserved disulfide bonds and N-linked glycosylation sites.

  • When performing IHC-P, including an antigen retrieval step is recommended to expose epitopes in formalin-fixed samples. Testing dilution series is advised.
