Biological Background: KLHL15 Function and Localization
- KLHL15 (Kelch-like protein 15), encoded by the gene KLHL15 (also known as KIAA1677), functions as a substrate-specific adapter for the CUL3 E3 ubiquitin-protein ligase complex. Related references: PMID:14528312, PMID:27561354, PMID:35219381
- It targets the DNA-end resection factor RBBP8/CtIP for ubiquitination and subsequent proteasomal degradation. This regulation favors error-prone non-homologous end joining (NHEJ) over error-free homologous recombination (HR) in DNA double-strand break repair. Related references: PMID:27561354, PMID:35219381
- KLHL15 also mediates ubiquitination and degradation of the serine/threonine-protein phosphatase 2A (PP2A) regulatory subunit PPP2R5B, modulating PP2A complex composition. Related references: PMID:23135275
- The protein localizes predominantly to the nucleus, consistent with its role in DNA damage response and protein turnover within this compartment.
- As a member of the Kelch-like protein family, KLHL15 contains characteristic Kelch repeat motifs.
- Mutations in KLHL15 are associated with X-linked intellectual disability (XLID103), highlighting its importance in neurological development.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–300) of human KLHL15, which includes the BTB domain and part of the Kelch repeat domain. Consider this when designing experiments to detect full-length or truncated KLHL15.
- For Western blot, the predicted molecular weight is ~70 kDa; use appropriate gel percentage and transfer conditions. Validate in human, mouse, or rat cell lines. For ELISA, optimize coating antigen and antibody dilution based on your assay format.
- This polyclonal antibody detects KLHL15 from human, mouse, and rat samples. Verify reactivity in your specific model system and consider sequence alignment for other species if needed.
- As a rabbit polyclonal antibody, it may recognize multiple epitopes; take this into account for applications requiring high epitope specificity.
