Biological Background: KLF7 Function and Localization
- Krueppel-like factor 7 (KLF7), also known as ubiquitous krueppel-like factor (UKLF), is a zinc-finger transcription factor encoded by the KLF7 gene (GeneID: 8609).
- KLF7 acts as a transcriptional activator (PubMed:16339272, PubMed:9774444). Related references: PMID:16339272 PMID:9774444
- It plays a critical role in neuronal morphogenesis and survival of sensory neurons (By similarity).
- KLF7 represses corneal epithelium differentiation (PubMed:28916725). Related references: PMID:28916725
- It functions as a metabolic regulator by modulating insulin sensitivity in pancreatic beta cells and skeletal muscle cells (PubMed:16339272). Related references: PMID:16339272
- KLF7 inhibits transcriptional inducers of adipogenesis and represses the expression of several adipokines, including leptin (PubMed:16339272). Related references: PMID:16339272
- The protein is localized in the nucleus.
- KLF7 is widely expressed, and alternative splicing generates multiple isoforms (Keywords: Alternative splicing).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a recombinant fusion protein covering amino acids 51–230 of human KLF7 (NP_001257871.1), a region upstream of the zinc-finger domains. This design may enable detection of KLF7 isoforms that lack the zinc-finger region; however, confirmation is recommended.
- For Western blotting, a band of approximately 33 kDa is predicted. Due to potential post-translational modifications or alternative splicing, additional bands may be observed. Include appropriate positive and negative controls to validate specificity.
- This antibody has been reported for use in ELISA; optimal coating concentrations and detection conditions should be determined empirically.
- Cross-reactivity has been demonstrated for human and mouse samples; for other species, consider sequence homology before testing.
