Biological Background: KLF4 Function and Localization
- KLF4 (Krueppel-like factor 4), also known as EZF or GKLF, is a transcription factor that can act both as an activator and repressor, binding the 5'-CACCC-3' core sequence.
- It plays a critical role in maintaining embryonic stem cell pluripotency and preventing their differentiation.
- Essential for establishing the barrier function of the skin and for postnatal maturation and maintenance of the ocular surface.
- Involved in the differentiation of epithelial cells and may also function in skeletal and kidney development.
- Contributes to the down-regulation of p53/TP53 transcription.
- Predominantly localizes to the nucleus, but can also be found in the cytoplasm.
- Contains C2H2-type zinc-finger domains and is subject to phosphorylation and ubiquitin-like conjugation.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 1-115 (N-terminal region) of KLF4, so the antibody may detect full-length and N-terminal isoforms. Consider validating isoform specificity if needed.
- For Western blot (WB), the expected molecular weight is approximately 55 kDa (UniProt calculated 54.7 kDa). Use appropriate loading controls.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and blocking conditions. Validate staining in known positive and negative tissues/cells.
