Biological Background: KLF4 Function and Localization
- KLF4 (Krueppel-like factor 4), also known as Epithelial zinc finger protein EZF or Gut-enriched krueppel-like factor, is a transcription factor that can function as both an activator and repressor. It binds to the 5'-CACCC-3' core DNA sequence.
- Plays a critical role in regulating the expression of key transcription factors during embryonic development and is essential for maintaining embryonic stem cell pluripotency while preventing differentiation.
- Required for establishing the barrier function of the skin and for postnatal maturation and maintenance of the ocular surface; involved in differentiation of epithelial cells and may also function in skeletal and kidney development.
- Contributes to the down-regulation of p53/TP53 transcription, linking it to cell cycle control and apoptosis pathways.
- Subcellularly localizes to both the nucleus and cytoplasm, consistent with its transcriptional regulatory functions and possible extra-nuclear roles.
- Contains zinc-finger DNA-binding domains, enabling direct interaction with DNA, and is subject to post-translational modifications including phosphorylation, ubiquitination, and alternative splicing, as suggested by proteomics data and structural studies.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning amino acids 180-479, which includes the C-terminal zinc-finger domains critical for DNA binding. This design may help detect both full-length and truncated isoforms if present.
- For Western blot, consider using a denaturing gel with appropriate protein loading controls and validate the observed band (~55 kDa) against known KLF4 expression in human cell lines or tissues.
- In ELISA, titrate the antibody against the recombinant immunogen or cell lysates to establish optimal working concentrations, as polyclonal preparations can vary between lots.
- Given the antibody's reactivity is listed for human, and KLF4 is widely expressed in epithelial cells, consider using human skin or intestinal epithelial cell lysates as positive controls.
