Biological Background: KLF15 Function and Localization

  • KLF15 (Krueppel-like factor 15), also known as kidney-enriched krueppel-like factor (KKLF), is a zinc-finger transcription factor that binds to the GA element of gene promoters.
  • Acts as a transcriptional repressor of CCN2 (connective tissue growth factor) expression, thereby playing a critical role in the control of cardiac fibrosis.
  • Inhibits cardiac hypertrophy by repressing the activity of key pro-hypertrophic transcription factors MEF2A and GATA4.
  • Regulates circadian expression of KCNIP2 in the heart by direct promoter binding, contributing to cardiac electrophysiology.
  • Involved in podocyte differentiation, indicating a role in kidney development and function.
  • Functions as a negative regulator of TP53 acetylation and inhibits NF-kappa-B activation by repressing EP300-dependent RELA acetylation.
  • Localized to the nucleus, consistent with its function as a DNA-binding transcriptional regulator.
  • Highly expressed in liver, skeletal muscle, and kidney; also found in cardiomyocytes, but expression is reduced in certain cardiac and glomerular diseases.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant protein spanning the N‑terminus (aa 1‑260) of human KLF15. Consider using full‑length positive controls to confirm detection of endogenous protein.
  • For IF/ICC, optimize fixation and permeabilization conditions to ensure nuclear staining; include appropriate isotype controls.
  • In ELISA applications, the antibody may be used as a capture or detection reagent; titrate and validate in the relevant sample matrix.
  • Due to the polyclonal nature, lot‑to‑lot consistency should be verified for critical quantitative assays.
  • Cross‑reactivity is reported for human samples; reactivity with other species is not guaranteed and should be independently tested if needed.
