Biological Background: KLF13 Function and Localization

  • KLF13, also known as BTEB3, RFLAT-1, and NSLP1, is a member of the Krueppel-like factor (KLF) family of zinc-finger transcription factors.
  • It functions as both an activator and repressor of gene transcription. KLF13 activates GC-rich minimal promoters in erythroid lineage cells (By similarity) and induces RANTES (CCL5) expression in T-cells (PubMed:17513757). Related references: PMID:17513757
  • As a repressor, KLF13 binds to the BTE site, a GC-rich DNA element, competing with the activator SP1, and additionally represses transcription by interacting with the corepressor Sin3A and HDAC1 (PubMed:11477107). Related references: PMID:11477107
  • KLF13 localizes to the nucleus, consistent with its role as a DNA-binding transcription factor.
  • It is a phosphoprotein and contains zinc-finger motifs that mediate DNA binding and metal coordination.

  • In Western blot (WB), a band migrating near 31 kDa is anticipated, consistent with the predicted molecular weight. We recommend using human, mouse, or rat whole-cell lysates as positive controls given the ubiquitous expression of KLF13.
  • For ELISA applications, users should optimize coating conditions and antibody concentrations; cross-reactivity with mouse and rat KLF13 orthologs may be leveraged for multi-species studies.
