Biological Background: KLF1 Function and Localization
- KLF1 (Krueppel-like factor 1), also designated EKLF, is a transcription factor crucial for erythroid development and fetal-to-adult globin switching.
- It binds to the CACCC box in the beta-globin gene promoter, activating beta-globin expression, and also stimulates BCL11A, which represses fetal hemoglobin genes (HBG1/HBG2), ensuring low fetal hemoglobin in adults.
- KLF1 activates the CD44 and AQP1 promoters (PubMed:21055716). Related references: PMID:21055716
- Upon sumoylation, KLF1 switches to a transcriptional repressor, interacting with CDH2/MI2beta and inhibiting megakaryocytic differentiation.
- The protein localizes to the nucleus.
- Expression is restricted to adult bone marrow and fetal liver, absent in myeloid and lymphoid cell lines.

- The immunogen is a synthetic peptide corresponding to the N-terminal region (aa 1–100) of human KLF1. For specificity verification, consider using peptide blocking assays or comparing reactivity against truncated constructs.
- For Western blotting, a band at approximately 38 kDa is expected. Validate using positive control lysates from erythroid cell lines or bone marrow extracts, and consider using reducing conditions.
- ELISA application may benefit from using the immunogen peptide as a coating antigen; titer optimization is recommended.
- This antibody shows cross-reactivity with rat KLF1. Validate in rat tissue samples where KLF1 expression is expected, such as bone marrow or fetal liver.
