Biological Background: KISS1 Function and Localization
- KISS1 encodes the precursor protein kisspeptin-1, which is proteolytically processed into several biologically active peptides, including kisspeptin-54, kisspeptin-14, kisspeptin-13, and kisspeptin-10.
- Kisspeptins are endogenous ligands for the G protein-coupled receptor KISS1R/GPR54, activating G(q)-dependent phospholipase C signaling that increases intracellular calcium and depolarizes GnRH neurons. Experimental Guidance and Technical Tips
- For Western blotting, a band near the predicted 15 kDa may be observed; however, potential glycosylation or precursor processing could produce additional bands. Validate using placenta or testis lysates as positive controls.
- In immunohistochemistry (IHC‑P), antigen retrieval may be required for this secreted target. Use placental tissue sections as a positive control and optimize blocking to reduce background.
- For ELISA applications, the antibody may be used for capture or detection; test peptide specificity and cross-reactivity with other kisspeptin isoforms using recombinant proteins.
