Biological Background: KIR2DL4 Function and Localization
- KIR2DL4 functions as a receptor for non-classical MHC class Ib molecule HLA-G when complexed with B2M and nonamer self-peptide.
- In decidual natural killer (NK) cells, KIR2DL4 binds peptide-bound HLA-G-B2M, triggering a senescence-associated secretory phenotype that acts as a molecular switch to promote vascular remodeling and fetal growth during early pregnancy. Related references: PMID:16366734, PMID:23184984, PMID:29262349
- KIR2DL4 signaling contributes to balancing tolerance and antiviral immunity at the maternal-fetal interface by regulating effector functions of NK cells, CD8+ T cells, and B cells. Related references: PMID:10190900, PMID:16366734
- Upon ligand engagement, KIR2DL4 initiates signaling from endosomal compartments, leading to activation of PRKDC-XRCC5 and AKT1, and ultimately NF-kappa-B-dependent pro-inflammatory responses. Related references: PMID:20179272
- KIR2DL4 is localized to the cell membrane and early endosome membrane.
- It is expressed in decidual NK cells, innate lymphoid cell type I (ILC1), and a subset of peripheral NK cells. Related references: PMID:29262349, PMID:19304799
- Belongs to the killer cell immunoglobulin-like receptor (KIR) family and contains Ig-like domains, a transmembrane helix, and is subject to alternative splicing and glycosylation.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (aa 22-242) of human KIR2DL4, potentially enabling recognition of both native and denatured protein; consider validating for WB and ELISA as listed.
- Predicted molecular weight is ~41 kDa; verify band specificity in appropriate positive controls (e.g., NK cell lines or decidual tissue lysates).
- Reported cross-reactivity with mouse and rat suggests epitope conservation; however, sequence alignment and experimental validation in these species are recommended.
