Biological Background: KIR2DL3 Function and Localization

  • KIR2DL3 encodes a killer cell immunoglobulin-like receptor (KIR) that belongs to the KIR family of inhibitory receptors expressed on natural killer (NK) cells.
  • It functions as an inhibitory receptor by binding specific HLA-C alleles (HLA-Cw1, HLA-Cw3, HLA-Cw7), delivering signals that prevent NK cell-mediated cytotoxicity and lysis of target cells.
  • The protein is a transmembrane receptor with two immunoglobulin-like domains, an ITIM motif in its cytoplasmic tail, and undergoes alternative splicing, resulting in multiple isoforms.
  • KIR2DL3 is localized to the cell membrane of NK cells and possibly other lymphocyte subsets.
  • Post-translational modifications include disulfide bond formation and N-linked glycosylation, which are critical for receptor structure and ligand binding.
  • KIR2DL3 is part of a diverse KIR gene cluster on chromosome 19q13.4; its expression contributes to NK cell education and self-tolerance.

Experimental Guidance and Technical Tips

  • The immunogen covers the extracellular domain (aa 22–245), so the antibody is expected to recognize the native receptor; consider optimizing epitope retrieval for formalin-fixed samples.
  • For Western blot (WB), a band near 38 kDa is anticipated; validate in relevant NK cell lines or primary cells under reducing conditions.
  • For immunofluorescence/ICC, fixation and permeabilization conditions should be empirically tested; paraformaldehyde or methanol can be suitable starting points.
  • For ELISA, the antibody may be used to detect recombinant KIR2DL3; include negative controls with irrelevant IgG to assess specificity.
