Biological Background: Kininogen 1 Function and Localization
- Kininogen 1, also known as alpha-2-thiol proteinase inhibitor or Fitzgerald factor, is a secreted protein that exists as high molecular weight (HMW) and low molecular weight (LMW) isoforms through alternative splicing.
- As an inhibitor of thiol proteases, kininogen 1 modulates proteolytic activity in plasma and plays a crucial role in blood coagulation by positioning prekallikrein and factor XI near factor XII.
- The HMW kininogen isoform inhibits thrombin- and plasmin-induced thrombocyte aggregation, while the LMW isoform inhibits thrombocyte aggregation but is not involved in blood clotting.
- The active peptide bradykinin is liberated from HMW kininogen and acts as a potent vasodilator with multiple physiological effects, including smooth muscle contraction, hypotension, natriuresis, diuresis, decreased blood glucose, inflammation mediation, and cardioprotection.
- Kininogen 1 is predominantly secreted into the extracellular space and plasma; it is detected at elevated levels in malignant ovarian, colon, and breast carcinomas.
- Post-translational modifications include glycosylation, hydroxylation, phosphorylation, disulfide bond formation, and pyrrolidone carboxylic acid formation, which may regulate its stability and function.
- The protein has been associated with hemostasis, inflammatory response, vasoactivity, and disease variants linked to coagulation disorders.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 148-427 of human KNG1, covering regions common to both isoforms; verify antibody reactivity with recombinant HMW and LMW kininogen if isoform-specific detection is required.
- Validate the antibody in relevant sample types, such as human plasma or serum, or cell lines known to express KNG1 (e.g., liver-derived cells).
- For Western blot, use appropriate reducing conditions to resolve the ~72 kDa full-length protein; note that some truncated forms may appear due to protease cleavage.
- In ELISA, consider pairing with a capture antibody targeting a distinct epitope to avoid competition and enhance signal.
- Cross-reactivity with mouse and rat has been demonstrated, but confirm in your specific experimental system as sequence divergence may affect binding.
