- KIFAP3, also known as Smg GDS-associated protein (SMAP), is a kinesin-associated protein involved in chromosome tethering to the spindle pole and chromosome movement during mitosis.
- It binds to the tail domain of the KIF3A/KIF3B heterodimer, forming a heterotrimeric KIF3 complex that may regulate the membrane binding of this motor complex (By similarity).
- The gene KIFAP3 (also designated KIF3AP) is mapped to human chromosome region and encodes a protein of approximately 91 kDa.
- According to UniProt annotations, the protein undergoes alternative splicing and is classified as a phosphoprotein; it contains repeat domains of unknown function.
- Subcellularly, KIFAP3 localizes to the mitotic spindle, consistent with its role in chromosome dynamics.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–200) of human KIFAP3. This antibody likely recognizes full-length KIFAP3 and any N-terminal isoforms that retain this epitope.
- In Western blot, a band around 91 kDa is expected. Validate specificity using lysates from cell lines with known KIFAP3 expression and appropriate controls.
- For immunofluorescence/ICC, KIFAP3 localizes to the mitotic spindle. Optimize fixation and permeabilization conditions, as spindle-associated proteins can be sensitive to standard protocols.
- The antibody is reported to cross-react with mouse and rat KIFAP3. Given high sequence homology in the immunogen region, it is likely suitable for these species, but independent validation is recommended.
