Biological Background: KIF7 Function and Localization

  • Protein identity: Kinesin-like protein KIF7, also known as ACLS, AGBK, HLS2, MMEDF, JBTS12, UNQ340. It is a member of the kinesin motor protein family.
  • Essential for hedgehog signaling: acts both as a negative and positive regulator of the sonic hedgehog (Shh) and Indian hedgehog (Ihh) pathways, acting downstream of SMO through SUFU-dependent and -independent mechanisms. Related references: PMID:21633164
  • Subcellular localization: Predominantly found in the cilium and basal body; also present in the cytoplasm and associated with the cytoskeleton.
  • Tissue expression: Detected in embryonic stem cells, melanotic melanoma, Jurkat T-cells, and widely expressed in heart, lung, liver, kidney, testis, retina, placenta, pancreas, colon, small intestine, prostate, and thymus.
  • Key post-translational modifications and functional domains: Classified as a phosphoprotein, with ATP-binding, coiled-coil regions, and potential ubiquitin-like conjugation sites. Contains motor and cargo-binding domains.
  • Disease relevance: Associated with ciliopathies such as Bardet-Biedl syndrome, Joubert syndrome, and related disorders involving intellectual disability and obesity.

Experimental Guidance and Technical Tips

  • For Western blotting, an expected band around 151 kDa is observed. Due to the large size, consider using a low-percentage SDS-PAGE gel (e.g., 6-8%) and extended transfer times.
  • The immunogen sequence (amino acids 1044-1343) is specific to KIF7; however, cross-reactivity with other kinesin family members should be assessed in relevant samples.
  • ELISA protocols should be optimized with appropriate antigen coating concentrations and detection conditions; this antibody is suitable for capture or detection in sandwich ELISA setups.
  • Since cross-reactivity is reported only for human, validate performance in other species if testing orthologous samples.
