Biological Background: KIF4A Function and Localization
- KIF4A (Chromokinesin-A) is a member of the kinesin superfamily of motor proteins, containing ATP-binding and microtubule-binding domains, and is essential for mitotic chromosome segregation.
- KIF4A is an iron-sulfur (Fe-S) cluster-binding motor protein critical for chromosome segregation during mitosis. Related references: PMID:29848660
- During the metaphase-to-anaphase transition, KIF4A translocates PRC1 to the plus ends of interdigitating spindle microtubules, promoting central spindle midzone and midbody formation and successful cytokinesis. Related references: PMID:15297875, PMID:15625105
- May also participate in mitotic chromosomal positioning and bipolar spindle stabilization.
- Located in the nucleus matrix, cytoplasm, mitotic spindle, midbody, and chromosomes during cell division.
- Highly expressed in hematopoietic tissues including fetal liver, spleen, thymus, and adult thymus and bone marrow; lower expression in heart, testis, kidney, colon, and lung.
- Post-translationally modified by phosphorylation and ubiquitination (isopeptide bond and Ubl conjugation), regulating its activity and stability.

- The immunogen sequence (aa 870–1080) lies in the C‑terminal region, which may provide high specificity by avoiding conserved motor domains. Validate reactivity in human, mouse, and rat samples.
- For Western blot, expect a band at approximately 140 kDa; use mitotic cell lysates as positive controls and confirm with knockdown or overexpression.
- For IHC‑P and IF/ICC, optimize antigen retrieval and permeabilization; strong nuclear and spindle staining is expected in dividing cells.
- ELISA performance should be optimized by checkerboard titrations to determine optimal coating and detection conditions.
